茄子空间诱变效应及其AFLP 分析

摘 要:

为了研究茄子空间诱变效果, 比较空间诱变系与原始对照间的分子水平差异, 揭示空间诱变的分子效应, 对 3 个不同 茄子原始自交系(K1、L1、M1)及其对应空间诱变系(K2、L2、M2)生物学性状进行了比较研究, 并利用 AFLP 分子标记技术 进行了 DNA 指纹分析。结果表明, 与原始自交系相比, 3 个空间诱变系的生育期、株型、果形均未发生明显变化, 发生变 化的性状是平均单果重和种子千粒重。诱变系 M2 的平均单果重比其对照增加 16.82%, 差异极显著; 种子千粒重有降低趋 势, 其中 L2 比 L1 显著降低了 10.24%。48 对 AFLP 选择性扩增引物在 6 个自交系和诱变系间扩增得到 40 条可以揭示诱变系 与原始自交系间多态性的条带; 部分目标片段测序结果经BLAST 分析表明与已报道的高频突变相关DNA 区域或蛋白同源 性很高。可见, 空间诱变技术能够引起茄子的遗传变异, 而且这种变异是以 DNA 水平的变异为基础的。

Induced Effects of Space Flight on Eggplant (Solanum melongena L.) and AFLP Analysis on Mutants

Abstract:

In order to study the mutagenic effects of space flight on eggplant (Solanum melongena), the biological characteristics were compared and the molecular difference was analyzed by amplified fragment length polymorphism (AFLP) between 3 original lines (K1, L1 and M1) and their corresponding mutant lines (K2, L2 and M2). The result showed that, the growth periods, plant types and fruit shapes of the three mutant lines had no significant difference compared with the original lines, whereas the average weight per fruit and seed weight per thousand were varied significantly. The average fruit weight of mutant line M2 increased significantly by 16.67% than that of its original line M1, and the seed weight per thousand of L2 decreased significantly by 10.24% than that of L1. 40 polymorphic bands between original lines and mutant lines were amplified by 48 AFLP selective amplification primer combinations. By BLAST analysis, it was showed that the sequence of some target AFLP fragments were highly homologous to the reported DNA region or protein implicated in high frequency mutation. It revealed that space flight could create genetic variations in eggplant which were based on the variation of their DNAs.
